The V-750 UV-Visible Spectrophotometer optical design uses precision double-beam optics with variable spectral bandwidth. A high sensitivity PMT detector provides accurate and reproducible measurements for low to high concentration samples.

By controlling the high voltage applied to the PMT, the dynode feedback offers a wider dynamic range than is generally found on uv-visible spectrophotometers. For high resolution measurements, such as gas and vapor phase spectroscopy, the spectral bandwidth can be set as narrow as 0.1 nm. Special low stray light slit settings which optimize the light image on the monochromator provide increased linearity to up to 5 absorbance units.

Operating Systems

Two user interfaces are available. Spectra Manager™ Suite the innovative cross-platform spectroscopy software, compatible with Windows 7 Pro (32- and 64-bit), Windows 8.1 and Windows 10 operating systems.

For simple operation, the intelligent remote module (iRM) has a new look and feel, with a color LCD touch screen. Data can be downloaded for portability or further PC data processing using Spectra Analysis.

The V-700 Series of UV-visible spectrophotometers has a growing list of software applications for both Spectra Manager™ and for the iRM. If you require an application which you don’t see listed, please let us know as we may already have it or we can prepare an application designed specifically for your requirements.

The V-760 is typically used where higher lineardynamic range is required for measurement of most absorbance, transmittance or reflectance in the UV-visible region, including liquids, solids and with options for specular absolute and diffuse reflectance.

The V-760 is a high-resolution UV-Visible double-beam spectrophotometer with double monochromator for exceptional stray light and absorbance linearity, variable spectral bandwidth and a photomultiplier tube (PMT) detector. The V-760 UV-Visible spectrophotometer includes a double monochromator for exceptional resolution with extremely low stray light (0.00008 %) for more accurate measurement over the widest photometric range (up to 8AU). The V-760 provides excellent sensitivity for diffuse reflectance or transmittance of solid and liquid samples using an integrating sphere. For high resolution measurements, such as gas and vapor phase spectroscopy, the spectral bandwidth can be set as narrow as 0.1 nm. Extended photometric linearity up to 8 Abs enables measurement of highly absorbing samples and is also useful for measurements of optical material with transmission characteristics that approach 0 % T.

The V-770 offers the unique benefit of single monochromator for higher optical throughput compared to double monochromated systems. This is especially usefulfor diffuse and specular reflectance measurements in the NIR where signal-to-noise can be a problem.

System Features

A PC-controlled objective carousel can be used to select any of the available 10X, 16X or 32X cassegrain objectives in combination with a standard optical zoom feature to provide enhanced video imaging of the sample using a high-resolution CMOS camera. Options include binocular viewing, polarized observation and refractive objective lenses.

Wide Spectral Measurement Range

The microscope system includes wide-band cassegrain objectives for high throughput transmittance/reflectance measurements continuously from 200 to 2700 nm (MSV-5200).

Scanning Dispersive Optical System

The optical system uses a precision double-beam scanning monochromator, it includes all of the features of a research grade scanning UV-Visible/NIR spectrophotometer with selectable spectral band width for variable spectral resolution as well as selectable circular apertures and an adjustable rectangular aperture for sample area discrimination for measurement through the microscope.

Polarization Measurement

An automated Glan-Taylor polarizer system (standard) provides polarization measurements in combination with the optional automated polarization analyzer.

    For many routine sample analyses the FT/IR-4000 can be paired with the ATR-PRO ONE single bounce ATR with a choice of prisms including a high throughput monolithic diamond, Germanium (ideal for carbon-black samples) or ZnSe. Simple Operation

The FT/IR-4000 Series is controlled by Spectra Manager™ Suite cross-platform software. This includes a wide variety of standard functions such as spectra measurement, Quick-Start, QC Compare and quantitative analysis. Additional applications can be added, such as chemometrics (PLS, PCR, CLS etc.). The sample measurement screen can be fully customized according to user requirements and each customized screen and parameters can be saved for different users (User Adaptive Software).

Sequence
Information such as the instrument status, measurement parameters and the sequence of data acquisition are displayed.

Data analysis results such as spectral comparison and sample quantification are displayed and readily available.

Real-Time Monitoring
A real-time data processing function can be used to observe and verify the spectrum during measurements. Spectra stored in thumbnails can also be overlaid with the current spectrum during sample measurements. Self-diagnosis and Validation Self-diagnosis functions allow verification of the current instrument status.

Zoom
Target peak(s) and/or functional groups can be easily checked by a zoom function.

Thumbnail
The measured spectra can be stored as thumbnails within the thumbnail window. The thumbnails can be individually selected and viewed in Spectra view

Real-Time Data Processing

The data collection/processing sequence can be defined to include several data manipulation commands, activated as the spectral data is collected. The final results after all processing commands are completed are displayed as the final spectrum.

FTIR Spectrometers System Features

Interferometer

The interferometer is manufactured as a solid cast aluminum block  and is completely sealed. The FT/IR-4000 uses a 45 deg Michelson interferometer and the FT/IR-6000 uses a 28 degree Michelson interferometer.   • Nitrogen Purge and Evacuation

    A multi-zone N2 purge system is standard to minimize the interference from environmental gases and vapors on the sample spectrum (CO2 and H20 elimination algorithms are also provided).  The interferometer, sample chamber and detector areas can all be independently evacuated (useful for fast sample loading) to completely eliminate CO2 and H20 for the most sensitive sample measurements and as the moving mirror does not really on an air-bearing the vacuum can easily be maintained. The FT/IR-6000 FTIR spectrometer can also be used in a combination of vacuum and purge for even faster sample turn-around.
  • Emission Ports and Light Introduction

    The FT/IR-6000 has several options for emission port optics with a range of window materials for measurement of external light sources. These can be coupled with customizable collection optics to focus a range of sizes and power outputs of light sources into the interferometer.
  • Rapid Scan and Step Scan

System description

The microscope is exchanged in the same way as an ATR or transmission holder in the sample compartment, without an requirement for adjustment or optical alignment. The excellent throughput allows for highly sensitive measurements for samples as small as 20 microns. The IRT-1000 can be operated from the integrated touch panel and a PC using Spectra Manager™ Suite. Sample is observation is made with an integrated color CCD camera and a 5 inch TFT color LCD monitor and recorded using the imaging function in Spectra Manager™. System description

The IRT-5000 microscope is used with either the FT/IR-4000 or FT/IR-6000 Series spectrometers, combining both micro and macro FTIR measurement, offering the most advanced microscopy and imaging systems. The systems are typically used in the mid-IR for materials identification and forensic analysis or in the near- and far-IR for more fundamental research.

The IRT-5000 FTIR Microscope includes an automatic carousel with a choice of cassegrains and refractive elements for observation and measurement. A mid-band MCT detector is included (with option for a high sensitivity peltier cooled DLaTGS detector). Up to two detectors can be installed to expand the spectral range of the microscope. The unique IQ Mapping function can be used for Transmission/Reflectance or ATR mapping without requirement to move the sample stage. The 16 element MCT offers faster response with excellent signal to noise and wider wavelength range than a focal plane array detector. The data processing is done using fast on-board computing so that there is no delay in data being delivered to the imaging software. This make this linear array microscope very fast and accurate.

The IRT-7000 includes up to two detectors as standard; a 16-channel linear array detector for infrared sample imaging and a single-point MCT detector. Standard Configuration
532/785nm Lasers with Matching Edge Filters

The NRS-4500 offers several standard configurations including the typical 633nm single laser or 532/785nm laser combination with matching notch or edge filters, with an option for a third laser which can be mounted internally or externally. Observation of the laser spot ensures perfect alignment of the target sample position and provides an indication of surface roughness. All laser wavelengths are selected in the software and once selected, the optical system (including the laser) is automatically aligned for optimal throughput and resolution. Four software selectable gratings control the spectral range and resolution from 8000 to 100 cm-1 (8000 to 50 cm-1 as an option). The grating direct drive system includes a rotary encoder to ensure excellent wavelength reproducibility ±0.2cm-1.

Fluorescence Rejection
457nm Laser and Patented Fluorescence Rejection Algorithm

JASCO has developed new and novel (patented) mechanisms to deal with sample fluorescence. As with other Raman systems, we can utilize laser wavelengths of 785 nm and up to 1064 nm, but we have recently incorporated a 457 nm laser that offers significantly higher Raman signal, improved spatial resolution and much lower fluorescence for many different sample types. Selecting a different excitation laser wavelength is only one of the ways the NRS-4500 minimizes fluorescence interference. The Fluorescence Rejection algorithm (patented) included in the Spectra Manager™ can effectively remove or minimize fluorescence regardless of the laser wavelength used either at the time of measurement or during post-processing.

  • Flexible Sampling Options

    With a choice of refractive objectives, both micro and macro measurements are possible as well as options for long working distance objectives for heating/cooling stages and other sampling accessories. The NRS-4500 can be used with either a manual sample stage or the PC-controlled, automated XYZ confocal mapping stage. A fiber probe can be added for external measurement.

  • Powerful ‘UserAssist’ Control for Experienced Spectroscopists and New Users

    The ‘UserAssist’ software aids the user in setting up the NRS-4500 for sample measurement; a simple sequence guide takes you through setup and optimization of measurement parameters with helpful advice and tips, such as a warning if you have the laser intensity set too high. When each of the parameters has been set, the NRS-4500 automatically selects the laser and matching notch filter, the grating for the appropriate resolution, focuses on the sample and then the spectral measurement is performed.

  • ‘Simple Search’ Function

    The new ‘Simple Search’ function is used with the automated XYZ stage. A new algorithm developed by JASCO (patent pending) analyzes the microscopic image and automatically selects measurement position(s) based on size, contrast and/or color of the target material. The user simply clicks the measurement button to execute spectral measurements of the desired sample positions.

  • Chemical Image Identification and Functional Groups Registry

    To provide faster Raman image processing, the Imaging Analysis software includes a ‘Registry’ of possible functional groups or other relevant compound information based on peak height or area calculations. After peak height or area calculation is developed, it can be saved to the Registry for future analysis use. The registry includes the peak calculation information and a ‘label’ describing the relevant vibrational motion. Image maps can be developed from mapping data simply by clicking on the desired, registered calculation to obtain a false color image of the peak intensity data for the registered calculation.

A rigid honeycomb optical base is the foundation of the instrument ensuring alignment and stability. System description

The accessories for J-1000 series can measure a variety of samples, from liquids to films to solid-states. Temperature control systems can be coupled with multi-position cells to run thermal melts, providing researchers with CD and thermodynamic data sets for conformational and folding studies. Automated high-throughput CD can obtain measurements on up to 192 samples without user intervention, saving both time and money. Kinetic and protein dynamics studies can be performed with our dedicated stopped-flow systems that can measure both CD and fluorescence.

The wavelength range has also been significantly extended, allowing measurements to be obtained in both the vacuum-UV and NIR spectral regions using the standard PMT detector (163 – 950 nm) and an optional InGaAs detector (up to 1600 nm). Instrument performance

Enhanced Vacuum-UV Measurement

The innovative optical system of the J-1500 permits the measurement of a CD spectrum in the vacuum-UV region down to 163 nm. The enhanced light throughput and new digital lock-in technology now allows for a small signal to be detected in a very noisy environment. Additionally, the optimization of the nitrogen purge efficiency helps to reduce the amount of oxygen in the optical bench so it does not bury the sample signal.

These features ensure that CD spectra can be obtained from strongly absorbing and high signal-to-noise (S/N) samples across the spectrum and into the vacuum-UV region. The quality of spectral data obtained, including data obtained at shorter wavelengths, substantially improves the accuracy of protein secondary structure analysis.

Exceptional Stray-Light Rejection

The double polarizing prism monochromators of the J-1000 series result in stray light lower than 0.0003%, enabling the instruments to obtain high-quality CD data even under conditions with high absorbances

High Sensitivity and Wide Dynamic Range of PMT Detector.

The PMT detectors used in the JASCO system are specially selected for the highest sensitivity and lowest birefringence. The wide dynamic range of the PMT detector ensures that large variations in signal intensity can still be detected, even for highly absorbing samples. The result is superior linearity between concentration and absorbance compared to other types of detectors including solid-state APD and CCD detectors.

The gain on a PMT detector is much larger  than most detectors, and therefore the signal amplification is larger. Under conditions with adequate light intensity, this typically leads to a larger amount of noise and therefore a lower S/N ratio. However, under conditions where there is a small amount of light hitting the detector (i.e. far-UV region), the gain of the PMT allows for good signal amplification at very low noise, increasing the S/N ratio.

Rapid Scanning

High sensitivity combined with a 10,000 nm-per-minute maximum scan speed allows the J-1500 to measure samples quickly, increasing productivity in your lab. An additional benefit is the minimal time exposure of biological samples to the high-energy UV light, minimizing the risk of sample degradation. A mechanical shutter can also be programmed to open and close only during actual measurement to further avoid UV exposure.

System description

The InGaAs detector has a wavelength range from 900-2500 nm, while the PMT ranges from 163-960 nm. The automatic detector interchange allows researchers to obtain both far-UV and NIR measurements without having to manually switch detectors. The addition of a grating monochromator to the double-prism monochromator setup provides low stray light and high light intensity, generating CD signals with high S/N performance throughout a large spectral range.

NIR-CD applications vary from conformational and chirality studies of metal coordination complexes to tuning chiral properties of nanomaterials. Additionally, Magnetic Circular Dichroism (MCD) can be used in conjunction with NIR-CD to probe a chromophore’s electronic structure as well as to induce chirality in compounds.

Instrument performance

Enhanced Vacuum-UV Measurement

The innovative optical system of the J-1500 permits the measurement of a CD spectrum in the vacuum-UV region down to 163 nm. The enhanced light throughput and new digital lock-in technology now allows for a small signal to be detected in a very noisy environment. Additionally, the optimization of the nitrogen purge efficiency helps to reduce the amount of oxygen in the optical bench so it does not bury the sample signal.

These features ensure that CD spectra can be obtained from strongly absorbing and high signal-to-noise (S/N) samples across the spectrum and into the vacuum-UV region. The quality of spectral data obtained, including data obtained at shorter wavelengths, substantially improves the accuracy of protein secondary structure analysis.

Exceptional Stray-Light Rejection

The double polarizing prism monochromators of the J-1000 series result in stray light lower than 0.0003%, enabling the instruments to obtain high-quality CD data even under conditions with high absorbances

High Sensitivity and Wide Dynamic Range of PMT Detector.

The PMT detectors used in the JASCO system are specially selected for the highest sensitivity and lowest birefringence. The wide dynamic range of the PMT detector ensures that large variations in signal intensity can still be detected, even for highly absorbing samples. The result is superior linearity between concentration and absorbance compared to other types of detectors including solid-state APD and CCD detectors.

The gain on a PMT detector is much larger  than most detectors, and therefore the signal amplification is larger. Under conditions with adequate light intensity, this typically leads to a larger amount of noise and therefore a lower S/N ratio. However, under conditions where there is a small amount of light hitting the detector (i.e. far-UV region), the gain of the PMT allows for good signal amplification at very low noise, increasing the S/N ratio.

Rapid Scanning

High sensitivity combined with a 10,000 nm-per-minute maximum scan speed allows the J-1500 to measure samples quickly, increasing productivity in your lab. An additional benefit is the minimal time exposure of biological samples to the high-energy UV light, minimizing the risk of sample degradation. A mechanical shutter can also be programmed to open and close only during actual measurement to further avoid UV exposure.

Highest S/N Performance

The high S/N (signal-to-noise) performance of the J-1000 series is achieved by a high throughput optical system and low-noise signal processing. RMS Noise < 0.004 mdeg (185 nm) for the J-1500 and J-1700, < 0.03 mdeg (200 nm) for J-1100.

The high-throughput circular dichroism (HTCD) measurement system has been developed for use with either the J-1500 or  J-1700 CD spectropolarimeter. This system can accommodate two 96-well plates (up to 192 samples) with protection at controlled temperature prior to measurement.

System description

Measurements using predetermined parameters for automated spectral scanning  and/or temperature ramping measurement can be made with a Peltier thermostatted flow cell. In addition to CD, LD and absorbance data, fluorescence can be measured simultaneously (with optional accessory).  Small sample amounts are required and can be recovered back to the micro-plate after measurement. The post-measurement analysis program allows batch processing of data and includes spectral correlation for quality control, determination of melting parameters and secondary structure estimation.

  • Optimized Optics

    The FVS-6000 optical system uses a 28 deg interferometer for high throughput and to minimize polarization effects with reflective optics to eliminate artifacts caused by birefringence, resulting in very high-quality VCD data.
  • Thermal Stability

    Thermal stability is maintained for long-term baseline stability by isolating the IR light source (high intensity ceramic light source), with thermal control of the photoelastic modulator and detector, which are all required for long duration measurements.
  • Wide Spectral Range

    The FVS-6000 uses a high sensitivity MCT detector optimized for VCD measurement across a wide spectral range from 3,200-850 cm-1. Optional detectors and filters can be added to extend the range to 4,000-750 cm-1, for measurement of OH and NH bands.     Two options for operation are available including Spectra Manager Suite innovative cross-platform spectroscopy software for Windows. And the intelligent remote module (iRM) with color LCD touch screen. These two control and analysis interfaces allow full instrument control and data processing. For regulatory compliance 21 CFR part 11 versions for both Spectra Manager and the iRM are  available as options.

  • High Speed, Precision and Accuracy

    For fast measurement, the P-2000 has a response speed of up to 6°per second, with a resolution of 0.0001°. The wide dynamic range from 0.001°  to ±90° enables measurement over a very broad range of concentrations. The sample chamber includes temperature measurement with the actual temperature constantly updated and displayed on the control screen.
  • Accessories

    To support a diverse range of sample types and measurements , the accessories and cells include many different options: a range of cylindrical and  demountable stainless steel cells, a Peltier thermostatted cell holder and monochromator options.     The automated validation program is compliant with the requirements of  GLP/cGMP, USP and  FDA.  Validation is used for automated periodic testing of light source energy, zero repeatability, and rotational accuracy/repeatability. Data acquired from the instrument is automatically transferred to the Spectra Analysis application for further analysis and reporting. Each instrument driver includes comprehensive hardware diagnostics and validation.

  • Spectra Manager™ CFR

    Spectra Manager™ CFR provides secure access and features compliant with 21 CFR Part 11; system access requires a user name and password which are assigned by the Workgroup Manager and individual Access Levels determine the access to Administrative Tools which includes instrument configuration, analysis applications, user setup, workgroup setup and security policies as well as system and application history logs. Three levels of electronic signatures are required including creation, review, and approval stages. An audit trail is recorded in each data file recording any data processing performed on the spectral data.

    System access levels for Administrator, Power User, Limited User and User are defined.
  • Electronic Signatures

    There are three levels of electronic signatures: Creation, Review and Approval. Electronic signatures are applied to spectral data files, Canvas templates or documents, instrument parameters and analysis methods.

    The system and application history are automatically recorded.

    Audit trails are recorded for every data file, recording data processing for all spectral data. Records are also made in the audit trail for instrument parameters, Canvas templates and Method files.
